چکیده

The localization of various metabolic processes of cells in particulate and soluble intracellular components has been studied intensively (l-3). The effect of added metals on these processes has also received attention (4-l3), but information on the metal content of the cell fractions is limited (14-16). The association of metals with proteins has been divided into two categories on the basis of their properties. Metalloproteins, wherein the metal is combined firmly with the protein, can be purified as stable, chemical entities (17). Metal-protein complexes, in which the metal is loosely bound, are subject to characterization only with difficulty. Any one metal in tissue may simultaneously be a component of one or more metalloproteins and metal-protein complexes, as well as being present in ionic form. The multiple role of metals has complicated the interpretation of analytical data from whole tissues. The investigation of metal content of subcellular fractions obtained by differential centrifugation (18-21) assists in further delineation of the problem. Reproducible and characteristic patterns of metal concentration, if demonstrated in any fraction, provide presumptive evidence for the functional significance of the metals found. In the present study, emission spectrogra, phy has been adapted to the simultaneous determination of several metals in fractions from livers of normal rats. It has been found that each metal is reproducibly distributed among the fractions, and that each fraction shows a characteristic and significantly different pattern of metal concentration.

MICROSOME-MEDIATED BENZO[A]PYRENE-DNA BINDING AND INHIBITION BY CYTOSOLIC FRACTIONS FROM LIVER AND SKIN OF ADULT AND WEANLING RATS

Biotransformation of benzo[a]pyrene (BaP) in the presence of microsomal fractions derived from liver and epiderm of adult and weanling rats was examined. The aim of this study was to evaluate the effect of age on the capacity of two organs in transformation of BaP. Subcellular fractions were prepared from skin and liver by ultracentrifugation and were used as the source of BaP metabolizing enzy...

Immuno-electron-microscopic studies on the subcellular distribution of rat liver epoxide hydrolase and the effect of phenobarbitone and 2-acetamidofluorene treatment.

The distribution of rat liver epoxide hydrolase in various subcellular fractions was investigated by immuno-electron-microscopy. Ferritin-linked monospecific anti-(epoxide hydrolase) immunoglobulins bound specifically to the cytoplasmic surfaces of total microsomal preparations and smooth and rough microsomal fractions as well as the nuclear envelope. Specific binding was not observed when the ...

Immunoaffinity purification, partial sequence, and subcellular localization of rat liver phospholipase A2.

Monoclonal antibodies against rat liver mitochondrial phospholipase A2 were used to develop a rapid immunoaffinity chromatography for enzyme purification. The purified enzyme showed a single band upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The sequence of the N-terminal 24 amino acids was determined. This part of the sequence showed only 25% homology with that of rat pancrea...

Inhibition of Microsome-Mediated Binding of Benzo (Α) Pyrene to "Dna By Cytosolic Reaction From Liver And Skin Rats in Cvitro

Purpose: The aim of this study was to evaluate the effect of age on the capacity of liver and epiderm of adult and weanging rats in transformation of Benzo (α) Pyrene. Materials and Methods: In a metabolic activiation assay system, cytochorome P-50 (from microsomal fraction) catalyses the formation of reactive epoxide of BaP which can then interact with exogenous DNA The capacity of cytochrome...

Rat liver adenosine triphosphate: adenosine monophosphate phosphotransferase activity. II. Subcellular localization of adenylate kinase isozymes.

Adenylate kinase activity (ATP:AMP-phosphotransferase, EC 2.7.4.3) was studied in subcellular extracts of rat liver tissue. Essentially all of the activity, over 150 units per g of tissue, wet weight (1.7 units per mg of protein), was released into the cytosol when the tissue was homogenized in 0.025 M sucrose. Isotonic (0.25 M sucrose) homogenization and separation of subcellular fractions by ...
